Acyclovir is a synthetic guanine nucleoside analogue with potent antiviral activity against herpesviruses.
It is selectively phosphorylated by viral thymidine kinase, then further converted to acyclovir triphosphate in infected cells.
Acyclovir triphosphate inhibits viral DNA polymerase and causes premature termination of viral DNA chain elongation.
It is primarily used in the treatment of Herpes simplex virus (HSV-1, HSV-2) infections and Varicella-zoster virus (VZV).
Clinically, it is indicated for genital herpes, herpes labialis, shingles (herpes zoster), and prophylaxis in immunocompromised patients.
